ABSTRACT

AIM To explore the inhibition in in vitro p roliferation human prostate cancer PC 3 cells induced by melatonin and the effe ct of combination of melatonin and 5-Fu. METHODS Cell growth cu rve, MTT assay, and acid phosphatase(ACP)activity were used to determine cell proliferation. Coomassie brillient blue assay and modified diphenylamine assay w ere used to measure the content of protein and DNA in PC 3 cells. ACP activity was measured by modified phenyl phosphat e method of kind and king and modified method of lowry. Co-administration effec t of MLT and 5-fluorouracil(5-Fu)on PC 3 cell proliferation was determined by MTT assay. RESULTS The IC 50 of MLT for inhibiting PC 3, CoLo205, K562, and HeLa cell proliferation was(1 20?0 30),(1 27?0 12) ,(1 60?0 16)and(2 25?0 11)mmol?L -1 , respectively. And MLT inhib ited all above cells proliferation in a concentration-dependent manner. The protein and DNA content and ACP activity in PC 3 cell treated with 2 mmol?L -1 MLT for 96 h were redu ced 58 77%, 54 97% and 89 24%, respectively. When PC 3 cells were treated wi th MLT in combination with 5-Fu 0 5 mg?L -1 , the inhibition rate enhance d. CONCLUSION MLT inhibited PC 3, CoLo205, K562, and HeLa cell p roliferation, and reduced the content of protein and DNA and ACP activity in PC 3 cell. MLT and 5-Fu co-administration enhanced the inhibition rate of PC 3.er.TheproteinandDNAcontentandACPactivityinPC3 celltreatedwith 2mmol?L-1MLTfor 96hwerereduced5 8 77% ,5 4 97%and 89 2 4% ,respectively .WhenPC3 cellsweretreatedwithMLTincombinationwith5 Fu 0 5mg?L-1,theinhibitionrateenhanced .CONCLUSION MLTinhibitedPC3 ,CoLo2 0 5 ,K5 62 ,

ABSTRACT

The respiratory route is an important approach for local and systemic delivery of drugs.The inhalation delivery of drugs to lung becomes a method when testing the efficacy and safety of these inhaled therapeutics in animal models,because of higher therapeutic index,lower side effect,and increasing bioavailability in inhalation compared with oral administration,as well as the low dose and high delivery efficiency.Recently,special devices were developed for inhaled drugs used for studies of pharmacodynamics and safety pharmacology in small or large animals,including mouse,rat,guinea pig,dog,monkey and sheep.These new developed equipments and methods have many excellences.This paper reviewed the progress of methodology for inhaled drugs tested in small and large animals.

ABSTRACT

Spontaneous locomotor activity is a widely used evaluation method in safety pharmacology of central nerve system and investigation of neuroscience.Spontaneous locomotor activity test is an important method that can investigate several animals' species and their associated indexes of spontaneous locomotor activity;it can also provide parameters for several pharmacological objectives including safety pharmacology of central nerve system.The new developed equipments and methods have many merits.This paper reviews the progress of methodology of spontaneous locomotor activity test in rats,pigs and monkeys.

ABSTRACT

AIM: To observe the effect and mechanism of Chinese traditional medicine JianNao mixture on function of learning and memory in memory disorder mice. METHODS: Step down test, step through test and "Y" maze test were used to examine the function of learning and memory in memory disorder mice induced by scopolamine, sodium nitrite and alcohol, and in dementia model mice induced by D galactose and aluminum chloride. Cholinesterase activity in brain, LPO and SOD in blood were measured. RESULTS: JianNao mixture could promote both the learning and memory function in impaired acquisition, consolidation and retrieval of memory in memory disorder mice induced by scopolamine, sodium nitrite and alcohol, respectively. It also could reduce wrong times and prolong latency in mice's step through test, depress cholinesterase activity of brain in aluminum chloride model mice, reduce wrong times in mice's "Y" maze test, enhance SOD activity of whole blood and decrease LPO content of brain in D galactose model mice. CONCLUSION: JianNao mixture can improve the function of learning and memory in memory disorder mice. The mechanism may be due to improve the cholinergic function and the balance of free radical in vivo.

ABSTRACT

Objective: To observe the pharmacological actions of Shenbao mixture on sex function in mice. Methods: Accessory sex organ weight and sperm activity in normal and model mice with kidney-yang deficiency induced by hydrocortisone were observed. The number of keratocytes,wet weight of the uterus and serum estradiol level in ovariectomized rats were examined. Sexual behavior and immune function were also observed. Results: Shenbao mixture can increase the accessory sex organ weight in normal and model mice, enhance the sperm activity and sexual behavior in normal mice and increase the number of vaginal keratocytes, wet weight of the uterus and serum estradiol level in ovariectomized rats. Shenbao mixture could also increase the wet weight of the thymus and the spleen and promote the proliferation of lymphocytes in model mice. Conclusion: Shenbao mixture can improve the sex function of mice.

ABSTRACT

The effect of bee pollen on serum IgM and IgG concentration was investigated in normal and immunodepressed mice.The levels of serum IgM and IgG in normal NIH mice were 0.80? 0.07mg/ml ( X ? SD ) and 11. 42 ?2. 31 mg/ml respectively. The concentration of IgM and IgG were all markedly lowered in immunodepres-ssed mice induced by cyclophosphamide 100 mg/kg/d sc for 4d,hydro-cortison 100 mg/kg/d sc for 6d,60Co ?-irradiation 600 rads and S-180 tumor-bearing in comparison to normal control, while IgG and IgM content in mice treated with bee pollen ( Pollen of Brassica campes-tris L. ) 10 g/kg/d orally for 1 month were significantly increased.The results showed that the stimulating effect of bee pollen on humoral immunity may be the base of beneficial effect in human being.

ABSTRACT

Preventive effect of pollen, (at 5~10g/kg/d orally for 1 month) on obe-sity and elevation of lipid in serum and liver was observed in obese mice induced by sub-cutaneous injection of L-monosodium glutamate (MSG)2 mg/g from 1st to 5th day afterbirth. The results showed that in MSG-treated mice the body weight and weight of fatin groin, around kidney, and in abdominal cavity and also the Lee's index were all redu-ced by pollen given orally before onset of obesity caused by MSG and during development.The total cholesterol and triglyceride levels in serum and liver were much higher in theMSG mice than normal mice. These results indicated that ora1 administration of pollencould prevent obesity of MSG treated mice.

Benign prostatic hyperplasia and prostate neoplasm were mainly disease which affect the living of the aging. Melatonin was a mainly hormone secreted by the pineal gland, which had a broad physiological effect. It was reported that melatonin affected the occurrence and development of the two diseases, meanwhile, the special melatonin binding site existed in the prostate tissue had been proved. This review focuses on recent progress in the studies about the aspects.

ABSTRACT

The effect of bee pollen of Brassica campestris L. and its alcohol extract on lipid peroxidation was observed in vivo and in vitro.The results showed that the production of lipid peroxides in normal liver hotnogenate of mice and elevation of production of lipid peroxides induced by cysteine and FeSO4 in homogenate were found to be inhibited significantly by in vitro addition of alcohol extract of bee pollen.The elevation of lipid peroxides in serum and liver in adult mice induced by alloxan 75 mg/kg(iv)or by administration of peroxidized corn oil 0.2 ml/mouse was markedly inhibited by oral administration of bee pollen (10 g? kg-1?d-1)for 20 days as compared with respective control groups.The level of lipid peroxide in geriatric mice was also markedly lowered by oral administration of bee pollen (10 g?kg-1?d-1)for 3 months as compared to non-treated geriatric mice.Based on the above in vitro and in vivo experimental results, it may be suggested that bee pollen and its alcohol extract protect tissues against destruction by lipid peroxides.

ABSTRACT

Immunoenhancement activity of bee pollen and its acetone extract was studied in normal, sarcoma-180 bearing, cyclophosphamide- and antilymphocyte serum-treated mice.Bee pollen and its acetone extract given orally for 30 days could significantly increase the production of serum anti-SRBC hemolysin (HC50) and the number of spleen plaque forming cells (PFC) in primary response to sheep red blood cell (SRBC) in young and adult mice. The acetone extract of bee pollen could significantly prevent the decrease of HC50) the number of PFC and specific rosette forming cells (SRFC), and the quantitative hemolysin of spleen cells (QHS) against SRBC in S-180 bearing, cyclophosphamide- and antilymphocyte serum-treated mice respectively.These results suggested that bee pollen of Brassica campestris L. and its acetone extract have immune-enhancement activity.
